In the draw made Monday, Portsmouth, the only Premier League side left in contention, come up against Championship side West Bromwich Albion.
The semi-finals will take place at Wembley Stadium on April 5 and 6.
A weekend of shock quarter-final results saw cup exits for both of last season's finalists, Chelsea and Manchester United.
It is 100 years since the semi-finals have only featured one top-flight side.
Portsmouth, who defeated Manchester United 1-0 Saturday, have not won the FA Cup since 1939, but they were semi-finalists in 1992 when Liverpool beat them on penalties.
In the weekend ties, Barnsley knocked out Chelsea 1-0, Cardiff beat top-flight side Middlesbrough, while West Brom defeated Bristol Rovers 5-1.
Welsh club Cardiff last lifted the trophy in 1927, the only time it has left England, and Barnsley have not done so since 1912. West Brom are five-time winners, most recently in 1968.
If any team other than Portsmouth lifts the trophy, it will be the first time since West Ham beat Arsenal 1-0 in 1980 that a team outside the top flight has won the competition.
